The church is located on the corner of Clarendon and Hotham Streets. The present church building was constructed in 1906. It replaced the 1864 Church building in George Street which was gutted by fire in 1905. The interior was repainted in 1987 in original Victorian colours. The building contains NZ Kauri pews, many memorials and a fine pipe organ. The parish hall at the rear was completed in 1888. It is one of few public buildings in East Melbourne, and is in constant use by community groups.

The original Vicarage was at 118 Hotham Street from 1868. The current residence in Clarendon Street was purchased in 1969. In August 1988 the Cairns Memorial Uniting Church was destroyed by fire and the two congregations amalgamated. Since 1863 the parish has had 14 Incumbents. The present one is Canon John Stewart who commenced his ministry here in February 2001. In addition to parish ministry, Fr John is continuing his former ministry at the Retreat House in Cheltenham by developing a centre for spirituality, retreats and spiritual direction.
