Wesley Uniting Church is situated at 148 Lonsdale Street. Wesley has two parishes; Wesley Central Parish Mission and the Chinese Parish. The present Wesley Church is the descendant of the earliest Wesleyan (Methodist) Church in Melbourne. That congregation first met in a simple chapel; on the corner of Flinders Lane and Swanston Street and later in a larger building on the corner of Collins and Queen Streets. It moved into the present blue-stone Gothic style building in 1858.

From the beginning the church was involved in relief work among the poor and needy. This work continues, with the Wesley central Parish Mission providing programs to assist a wide spectrum of the community, including; the aging, people with disabilities, people in crisis, and young people. ' Wesley Central Parish Mission's sole purpose is to be faithful to Almighty God by using its service, witness and worship to show God's love for all people as revealed through Jesus Christ.'
